His lab studies the mechanisms underlying brain state changes in awake mice using in vivo whole-cell patching and fluorescence imaging. This webinar will describe the experimental imaging effort for the IARPA-funded MICrONS project, combining large-scale anatomical connectivity and functional imaging data to understand computation in cortical circuits. This data set began with in vivo recordings of nearly 70,000 cells in a single 1 mm^3 of visual cortex using two-photon and three-photon calcium imaging. In this same animal, collaborators at the Allen Institute for Brain Science and Princeton performed EM imaging and dense 3D reconstruction of the same volume at single-synapse resolution. In the context of MICrONS, this massive functional/structural data set will be mined for computational principles to advance artificial intelligence. More generally, this experiment is an important step towards a future where detailed anatomy and in vivo recordings of large neuronal populations can be routinely collected in the same animal, enabling a better understanding of the mechanisms underlying the behavior of distributed neural circuits in both health and disease.
