Abstract:

Protein therapeutics is a fast-growing market. Ensuring product quality plus speed to market is crucial. We will review the key stages of our CLD platform to ensure the manufacturability of the biologics. By leveraging host cells growth characteristics and new technologies we have reduced our timelines and enhanced our titers.


Over the past two decades, recombinant therapeutic proteins have changed the face of modern medicine and our ability to treat, manage and prevent a wide variety of diseases from cancer to autoimmune disorders. Despite the abundance of different types of expression hosts (cell lines), nearly 70% of all recombinant protein therapeutics marketed today are made in Chinese Hamster Ovary (CHO) cells. Today, solutions for complex human diseases are being addressed with increasingly complex protein therapeutic modalities. These include molecules such as bi-specific antibodies, DARPins, triabodies, novel scaffolds decorated with peptides, enzymes and growth factors, to name a few. The pressures on the CHO expression system to express these complex proteins led Selexis to develop new modules for its technology platform that address a range of issues such as translation/secretion bottlenecks, lead candidate selection and, last but not least, quality assessment and barcoding of the clonal production cell line using next generation sequencing. At Selexis, we believe in the promise of these newer biologics and have committed significant resources to ensure our technologies can meet the new expression challenges that these non-natural proteins present.
