Visualizing the RNA World: In-Situ Hybridization for Bench and Clinic

“RNA In-Situ Hybridization for Research and Clinical Diagnostics: Everything Old is New Again.”

By: Yuling Luo, Ph.D., CEO Advanced Cell Diagnostics

Gene expression is traditionally analyzed by “grind-and-bind” assays such as RT-PCR. However, the resulting data lack critical cell-type specific and spatial information important for functional interpretation. Recently, we have developed a technology capable of detecting and quantifying single RNA molecules in situ, providing a powerful tool for single cell analysis of gene expression in research and clinical diagnostic applications.

“Quantitative In-Situ mRNA Measurement For Companion Diagnostics.”

By: Dr. David L. Rimm, M.D., Ph.D., Professor, Pathology, Yale University School of Medicine

New methods of in-situ hybridization using paired sets of primers or locked nucleic acids have made in situ measurement of mRNA reproducible and quantifiable.  Here we show some examples of the use of these tools, in combination with protein assessment, to predict outcome or response to therapy in cancer.

Before ACD, Dr. Luo has been a founder of Panomics (acquired by Affymetrix in 2008), and was one of the earliest senior scientists at Exelixis. He has authored many peer-reviewed publications in top-tier scientific journals and his inventions are the subject of over a dozen patents and patent pending applications. He received his doctorate in biochemistry from Case Western Reserve University and conducted post-doctoral research at the University of Pennsylvania.

Dr. David L. Rimm, M.D., Ph.D., Professor, Pathology, Yale University School of Medicine

Dr. David Rimm is Professor of Pathology, Director of Yale Pathology Tissue Services and Director of Translational Science in Pathology at the Yale University SOM.  He completed his MD-PhD at Johns Hopkins University Medical School, Pathology Residency at Yale and a Cytopathology Fellowship at the Medical College of Virginia. His research focuses on quantitative pathology using AQUA® technology invented in his lab.  He is an author of over 230 peer-reviewed papers and 8 patents and was the scientific co-founder of HistoRx, a digital pathology company and Metamark Genetics, a prognostic determinant company.
